Re: What is the absolute risk?

One figure I found particularly interesting was supplemental figure 4, which lists apoE levels, total cholesterol, LDL, HDL, etc by genotype. There is a clear positive correlation between E4 load and LDL and negative correlation between E4 load and HDL, although the differences aren't huge. One meas...

Re: Karin Michels talk on nutrition "errors"

Not understanding German, I have relied on summaries of the video. I found this to be the most even handed: https://www.vox.com/science-and-health/2018/8/23/17773494/coconut-oil-poison-saturated-fat . I think the Karin Michels video is a very strong response to the bulletproof coffee / Dr. Oz "...

Re: ApoE 2/4

57 years old is quite young for an Alzheimer's diagnosis, and the science around the impact of apoE on AD (either E2 or E4) before the age of 60 is a mixed bag. With a strong family history of Alzheimer's disease, particularly if the cases are before age 65, I would look at some of the familial AD g...

Re: E4: why no urgent push for structure correctors, gene editting?

One would want to increase lipidation of ApoE4, decrease apoE4 levels (preferentially non-lipidated apoE4), or alter apoE4 so that it could be more lipidated, or less toxic (Wang et al, 2018). I don't think these approaches need to be mutually exclusive either. The holy grail! I like this, Harrison...

Just curious if this event happened, and if so, what was presented.

Re: E4: why no urgent push for structure correctors, gene editting?

Hi Fiver, I independently came to a lot of the same conclusions that you have. Specifically, apoE4 is less lipidated by ABCA1 (Boehm-Cagan et al 2016) and that non-lipidated apoE is the problem (Liao et al, 2018). One would want to increase lipidation of ApoE4, decrease apoE4 levels (preferentially ...

Re: Simvastatin and the ApoE4 Allele

I actually couldn't find much actual evidence that the different statins penetrate the BBB differently. If someone has those studies I would love to hear about them - it makes logical sense, just haven't seen the data... ...The more I read the less clear it seems.... Ain't that the truth. To muddy ...

Re: Simvastatin and the ApoE4 Allele

Very interesting topic. Some of the comments suggest why it may be so difficult to see a consistent effect of statin use on AD: 1. Statins vary in their ability to cross the blood brain barrier among other properties (synthetic vs. fungus-derived; effects on lowering apoE levels!) 2. People may chan...

Re: A provocative critique of Banner's 4/4 RCT

So I suppose you are talking about the Generation Study 1 https://www.clinicaltrials.gov/ct2/show/NCT02565511 which does not screen for amyloid but does follow with PET scans. Yes, I was referring to the generation 1 trial. They can later stratify for those who are amyloid negative at the first sca...

Re: A provocative critique of Banner's 4/4 RCT

Interesting critique and response. It is interesting that a positive amyloid scan is not considered an eligibility criteria, and the investigators indicate that is a plus since E4/4's that are amyloid negative are at risk. However, they are only recruiting E4/4s that are 60 and older. Based on the w...
