Re: Both parents carry APOE4

I will borrow from a previous post on this: https://www.apoe4.info/forums/viewtopic.php?f=2&t=3238&p=38804#p38804 Using typical US/European apoE4 frequencies and knowing nothing about parents, I came up with the following chances for siblings of an apoE4/4 carrier (where X=E2 or E3): EX/X: 2...

Re: Erasing the damage caused by apoE4 in human genes? New study by Gladstone Institutes

Is it possible that when you chemically attach one molecule (structure corrector) to another molecule such as APOE4 that has functional interactions with over 1500 genetic locations in the human body, you simply cannot expect anything other than potentially massive and potentially dangerous side ef...

Re: Neuroenergetics provides new answers to Alzheimer's Disease

In looking at your paper, I did not see any mention of early onset AD, particularly the forms caused by autosomal dominant mutations such as Presenilin1 (PS1) or APP. Do you think that a neuroenergetic deficit precedes plaque accumulation in those patients as well, or do you believe that early onset...

Re: 23 and me surprises

Hi Chrissy, I am glad you are seeking confirmatory testing. My mom tested with 23andMe (v4 chip) and her promethease report suggested Lynch Syndrome. She is in her seventies and has not had any of the problems associated with it. Neither my brother or I who tested with the v2 and v3 chips respective...

Re: 3/4 with Good Health

I am E3/4. My LDL is considered borderline-high (140 mg/dL), but I have HDL around 79 and triglycerides around 40, and a coronary calcium scan score of 0. The best LDL measurements I ever had was eating low-fat, high-carb, but I weighed 20 pounds more and was hungry all the time, with low HDL and hi...

Re: 'Statin for the brain' vaccines in news today

Boy they sure didn't provide much information about the drug candidates, did they? Some seem to be AB antibodies, and similar, to slow aggregation. But I have no idea what a "statin for the brain" is. All sorts of mixed messages in that metaphor. Not great "science communication"...

Re: 3/4 with Good Health

Welcome Andru! 'I can comment on the Alzheimer's side of things. As we learn from all of the healthy E4/4s in this forum, APOE4 by itself does not "cause" AD. It should not be taken lightly, but there are things that can be done (which you are clearly doing!). E3/4 is half the risk of E4/4...

Re: 'Statin for the brain' vaccines in news today

The Daily Telegraph article is an interesting take on the current status of Alzheimer's disease drug development. Since all of the amyloid directed treatments have failed in patients with established disease, the push has been to treat earlier and earlier. With some of those same drugs now failing t...

Re: My Promethease: No wonder I am confused

It's unfortunate that Promethease has been causing so many problems with interpretation of E2/E4 vs E1/E3. The fact of the matter is that E2, E3, and E4 make up over 95% of ApoE genotypes, and it is highly, highly unlikely that someone would carry the E1 isoform. Regarding E2/E4 and AD risk, there i...

Re: Promethease

I am sure others will weigh in, but I wanted to give some general suggestions. First of all, 23andme is only giving you a partial look into your DNA. There have been multiple versions of the chip that 23andMe uses, and some have more genotypes than others. Beyond that, there is whole exome sequencin...
