Re: Rapamycin rescues E4 mice

Hi RJones, Dose has nothings to do with dose in mice. Mouse dose just shows humans need a low dose that would reduce mTOR 1; but not mTOR2. Mice very different rates of metabolism of rapamycin. Heres how get 6 mg once a week. 6 mg as single dose in humans has a significant physiologic effect. 5 mg d...

Re: Overview of the biggest threats

This type statistics are totally wrong and highly abusive to APOE4 community.

Re: Alzheimer's UK estimates APOE risk

These type statistic which discuss total dementia with AD included and general population with E4 are extremely misleading and very much underestimate risk for E4 carriers. Example is Diabetes. they give 6/100 at age 75 and 10/100 with diabetes. Looking at specific data (from Peila, Honolulu-Asia Ag...

Re: Rapamycin rescues E4 mice

Hi RJones, recalculating.
Mouse eats 5 gm =25 calories a day or 1/100 human calories. Mouse dose rapamycin @ .07 mg/day. Then human dose 7 mg day or 50 mg a week based upon calories consumed by body weight compared to mouse.

Re: Rapamycin rescues E4 mice

Hi RJones, Dose figured by body weight. The Harrison preparation was 14 mg/kg food. Calculation was average mouse was 30 gm and consumes 5 gm food a day. So they calculated mice consumed 2.24 mg/kg a day per mouse weight. Human weight is usually taken as 70kg (154 pounds). The equivalent human dose ...

Re: Rapamycin rescues E4 mice

Hi, As regards stage at which rapamycin rescues AD mice there are 3 critical papers: 2011: Oddo group, "Inducing autophagy by rapamycin BEFORE, but NOT AFTER, the formation of Plaques and Tangles Ameliorates Cognitive Deficits." Here when started rapamycin in advanced disease in 15 month o...

Re: Is it useful to add cannabinoids yet?

Note for Potheads: See "Cannabinoid modulation of hippocampal long-term memory is mediated by mTOR signaling", Puighermanal 2009. "Cognitive impairment is one of the most important negative consequences associated with cannabis consumption. The CB1 cannabinoid receptor activation acti...

Re: Interventions had OPPOSITE impact on lab results!

Hi jcosmo, Your lipid response is exactly what I would expect. You have 2 good reasons to increase lipids in blood, especially TG to go up. First, you are on high fat diet. Second you are on diet to lower mTOR. mTOR causes lipids to go into tissues and lowering mTOR increases lipids in blood. Decrea...

Hi Everybody, For starters, Is rapamycin an immune suppressant. See two papers, Mannick study (ref 51 and Blagosklonny Ref 43) and also my discussion on rapamycintherapy.com. Rapamycin was approved in 1999 as a biologic poison. Specifically to be used for transplant medicine. This required daily dos...

Re: APOE mentioned in Times today

The problem is revealed in above story. "Pfizer giving up on AD research". But Pfizer makes rapamycin. Rapamycin prevents AD in all mouse lab studies. Rapamycin is a generic drug. So Pfizer has no interest in Rapamycin. Pfizer wants to develop a new drug for which they could make 100 billi...
