Re: Homotaurine (tramiprosate) appears to be a structure corrector

If my nausea doesn't go away I'm going to do a bulk order of valiltramiprosate (valylamide prodrug of homotaurine) from a chemical supply house. Actually... no: I can't find a non–prohibitively high price. Since yesterday the nausea is mostly gone, fortunately (and that, despite having COVID and be...

Re: Homotaurine (tramiprosate) appears to be a structure corrector

Thanks Tincup. Yes, I saw the older thread. One of the things I cherish about this group are the detailed reports from people about their self-experimentation. I always take those reports into account when making my own health decisions. But I also look closely at published science. My take-away abo...

Re: Homotaurine (tramiprosate) appears to be a structure corrector

I just started three days ago. So I'm hoping that the nausea -- experienced by around 20-25% of the people who take homotaurine -- is just me adjusting, or caused by something else. My still somewhat cursory look at the literature (I'll be diving in more deeply soon) made me conclude that the best d...

Homotaurine (tramiprosate) appears to be a structure corrector

https://molecularneurodegeneration.biomedcentral.com/articles/10.1186/s13024-023-00620-9 This is amazing news. It would explain why trials show an allele-dose effect (homozygote > heterozygote > non-carrier). Although note that an older paper suggests that the molecule being corrected is Aβ42: https...

Re: Why Krill Oil Beats Fish Oil

Hi everyone. Thanks for the great ideas.

Salmon roe looks like a great choice. Be sure to check ingredients. A lot of salmon roe products contain sorbitol (again: this might not be a problem, but many of us may want to avoid sorbitol).

ChicagoGirl, like TeaandGlasses – and I suspect everyone here – I'm grateful for your taking the time to document your experience. Invaluable! And thank you to everyone else who also has made this such a useful and inspiring discussion. I want to note something in the event others try to join this t...

Re: New idea for gene therapy: peripheral apoE3

The gene therapy field is exploding with ideas. CRISPR-cas and other gene-editing techniques are under development for a number of neurodegenerative conditions, though I'm more hopeful about the work being done in David Liu's lab, which involves precisely changing one nucleotide ("letter")...

Re: New idea for gene therapy: peripheral apoE3

Julie, yes, showing a change in cognition is what we want to see. As far as Lexeo goes, I'm more hopeful about LX1020, which is still in development: https://www.lexeotx.com/programs/cns-programs/alzheimers/ LX1020 includes a cassette that knocks down apoE4 in the brain, which might be more importan...

Re: New idea for gene therapy: peripheral apoE3

They reported that 1) the therapy appears safe, and 2) apoE2 is being expressed (it's in the CSF).

I would say it's far too early to know whether it improves cognition, esp. given that the first cohort received such a low dose. But we should know more within a year or so.

New idea for gene therapy: peripheral apoE3

Hi everyone. I attended the recent Lexeo webinar, and was impressed with the progress Professor Crystal and his team have made. As we APOE-ε4 carriers wait for Lexeo's approach to come to market – which of course could take years – I was thinking of what other gene therapy approaches could be tried,...
