Ready for Quadruple Strength Aricept?

Post by J11 »

Results from the phase 2 trial for CPC-201 showed a 90% response rate over 26 weeks in those with moderate AD.
The smart idea that they came up with is to combine solifenacin with the Aricept.
Solifenacin does not enter the brain though it counteracts the side effects of Aricept in the rest of the body.

This report from a non-peer reviewed meeting abstract only reported a 90% benefit from subjective measures given by those who were not blinded to the treatment. Furthermore, there was no control, only a claim that control groups from other studies would imply a benefit from the new drug protocol. Nothing to get excited about until Phase 3.

The mean change in ADAS-Cog over the co ... 0 percent.

I suppose.

Yet, it is already understood that a dosage effect exists.
Until now no one had figured out how to unlock it.

There were 0 percent side effects which is quite a bit better than the current side effect profile.

"The primary endpoint of this Phase 2 single-blind, crossover trial was to safely increase the tolerated dose of donepezil from 10 mg/day up to as much as 40 mg/day. This trial consisted of 41 moderate (MMSE 10-20) Alzheimer’s type subjects who were being successfully treated with 10 mg/day of donepezil. Moreover, in the maintenance phase of the trial, subjects experienced significantly less dose-limiting side effects than those that would be predicted for 10 mg/day of donepezil. "

"Although this Phase 2 study was not designed nor powered to provide meaningful estimates of anti-dementia efficacy, both the ADAS-Cog and the CGI-I scales in the study yielded positive signals of enhanced efficacy. ...

“Pre-clinical studies have repeatedly shown that there is a strong dose/efficacy relationship associated with AChEIs. Further, PET scan studies in humans show that at the typical dose of 10 mg/day of donepezil, the central enzymatic inhibition is no greater than about 30 percent, substantially lower than that which would be expected from optimal dosing. Yet, historically, it has not been possible to tolerably increase the dose of donepezil to meaningfully higher levels,” "

Fairly impressive that this was in moderate AD.

Is Aricept considered effective in severe AD?
"Aricept is a prescription medicine to treat mild, moderate, and severe Alzheimer’s disease"
Looks like the solifenacin Aricpet combo will be a topic of discussion at the next doctor's appointment.

Would Quadruple strength Aricept be effective in severe AD?

“If successful in Phase 3, CPC-201 could be the new standard for the symptomatic treatment of Alzheimer’s disease and benefit millions of suffering patients.”
