epsilon 33, 34, 44 - whatever- what difference does it make????

Post by J11 »

Completely shocked!!

When Ancestry reports the genotypes for epsilon 2,3,4 they don't really mean the genotype stated. So,
when they say it is epsilon 33, it might actually mean epsilon 44. Does anyone on the forum feel that
epsilon 33 and epsilon 44 have essentially equal medical significance?

Ancestry.com has 8 million customers and is expecting to soon surpass 10 million.
Can you truly be THAT dumb and still be THAT successful?
The New Year is still young, though this has to be the stupidest thing of 2018 (I hope).
How could anyone top this?

https://www.snpedia.com/index.php/Gs246

snpedia tried to provide the context for the Ancestry results, though in this instance I
think it would be best to simply ignore the APOE epsilon genotype from Ancestry.com.
How does telling people that they might have epsilon 33 genotype when they are actually
epsilon 44 provide useful information?

Whoa.... Important find. So anyone who's learned their (supposed) APOE status through Ancestry.com must have it independently verified. Great info, J11.

Wow, maybe that should be pinned at the top of the New Here section.
ApoE 3/4 > Thanks in advance for any responses made to my posts.

circ here is the quote from snpedia. I have left it in its original because it is very difficult to believe.

"Word of caution to those with data from Ancestry.com: in our experience, based on data in OpenSNP and from Promethease users since 2006, Ancestry data always reports rs429358 as (T;T), even for people who's data from other sources indicates they are (C;T). Therefore, until Ancestry corrects this false negative problem, be aware that the ApoE genosets assigned by Promethease will be inaccurate when based on (inaccurate) Ancestry data. Specifically, some percentage of Ancestry users who are supposedly gs246 positive (Apo-ε3/ε3) are actually either gs141 (Apo-ε3/ε4) or gs216 (Apo-ε4/ε4)."

...since 2006, Ancestry data always reports rs429358 as (T;T), even for people who's data from other sources indicates they are (C;T).

So the Ancestry APOE genotype is like a clock that is always set at the same time. However, unlike a broken clock, miscalled genotypes would not be correct even once per day.

Yes, Whoa. Has this been resolved? Am I understanding this correctly? I have my kit here and have not yet registered or used it. Apoe4 status would be my primary reason for doing this. Should I get the money back and use someone else? These are old posts but rolled up under "unread" for me today.

Drae, if you were planning to use Ancestry for E4 testing, then I would request a refund. You can get a test for just ApoE4 through other labs, like Life Extension. Or use 23andme, if you want to look for other risk factors like MTHFR.

And note, I have updated the wiki on E4 testing to reflect the snpedia.com warning.

Julie, we need to update the thinking about testing page, too.

Noted, thanks. Although we don't recommend Ancestry.com per se in that article, I will make a note of warning under item #2.

Ancestry.com epsilon 4 genotyping is Totally Bogus

Post by J11 »

I think I need to make an executive decision here concerning the title of the thread:
"epsilon 33, 34, 44 - whatever- what difference does it make????"

The title does not clearly summarize what the thread is about.
Many of those even on our forum probably have no idea what it is about.

"Ancestry.com epsilon 4 genotyping is Totally Bogus"
would be more to the point. Anyone who might be able to give a better suggestion is certainly
encouraged to do so.

Oh, great... you really do need executive power to change the name of a thread.
I tried and it only changed my post.

Could anyone with the proper authority help out?
